ABSTRACT
One of the obstacles limiting progress in development effective cancer therapies is shortage preclinical models that capture dynamic nature tumor microenvironments. Interstitial flow strongly impacts response to chemotherapy; however, conventional vitro largely disregard this key feature. Here, a proof principle microfluidic platform for generation large arrays breast spheroids are grown under close-to-physiological biomimetic hydrogel reported. This spheroids-on-a-chip model used time- and labor-efficient studies effects drug dose supply rate on chemosensitivity spheroids. The capability grow from patient-derived cells different subtypes shown, correlation between vivo efficacy on-chip spheroid demonstrated. proposed can serve as an personalized screening new anticancer drugs.
