ABSTRACT
Abstract Tertiary lymphoid structures (TLSs) provide specialized niches for immune cells, resulting in improved prognoses patients undergoing cancer immunotherapy. Shaping TLS‐like may improve anti‐cancer immunity and overcome the current limitations of cell‐based Here, it is shown that stromal vascular fraction (SVF) from adipose tissues can enhance dendritic cell (DC)‐mediated T by inducing ectopic lymphocyte clusters. SVF cells expanded ex vivo have phenotypes functions similar to those fibroblastic reticular a secondary organ, their properties be modulated using three‐dimensional spheroid culture coculture with DCs spiked antigen‐loaded iron oxide–zinc oxide core‐shell nanoparticles. Thereby, combination spheroids mature significantly augments recruitment retention at injection site. This strategy elicits enhanced antigen‐specific response anti‐tumoral mice, illustrating potential novel immunotherapeutic design as structural scaffold TLS.
