An Off‐the‐Shelf Artificial Proregenerative Macrophage for Pressure Ulcer Treatment

ABSTRACT
Abstract Cell therapy is a promising approach in regenerative medicine. However, maintaining the survival and function of injected or implanted therapeutic cells remains substantial challenge to success. In vivo modulatory strategy for cell therapeutics has been recently developed, but suffers from limited efficacy injured tissue microenvironment with chronic inflammation. Here, an off‐the‐shelf artificial macrophage (artM) assembled by M2 macrophages‐derived lysate proteins‐loaded poly (lactic‐co‐glycolic acid) (PLGA) microspheres coated membrane developed. The synthetic artM fabricated batches maintains its bioactivity long‐term cryostorage. Significantly, recapitulates essential inflammation‐regulatory proregenerative characteristics endogenous macrophages, including initiating polarization, resolving excessive inflammation releasing anti‐inflammatory cytokines growth factors, neutralizing endotoxins proinflammatory cytokines, augmenting T‐helper 2 (T H 2) immune response, coordinating migration proliferation. mouse model deep pressure injury (DTPI), induces regeneration modulating inflammatory microenvironment, promoting angiogenesis, reducing scar deposition, accelerating renewal skin appendages. Depletion macrophages mice ulcers highlights immunomodulatory proangiogenic functions as effective autogenous macrophages. Collectively, engineered represents cell‐free, proreparative alternative wound management.
