ABSTRACT
Abstract We introduce a general method to transform antibodies into ratiometric, bioluminescent sensor proteins for the no‐wash quantification of analytes. Our approach is based on genetic fusion antibody fragments NanoLuc luciferase and SNAP‐tag, latter being labeled with synthetic fluorescent competitor antigen. Binding antigen, here drugs, by displaces tethered from disrupts resonance energy transfer (BRET) between fluorophore. The semisynthetic sensors display tunable response range (submicromolar submillimolar) large dynamic (Δ R max >500 %), they permit analytes through spotting samples onto paper followed analysis digital camera.
