ABSTRACT
Hepatic microcirculatory perturbation is observed after ischemia/reperfusion. Endothelin-1, a potent vasoconstrictive peptide, known to modulate local circulation. This study was designed examine whether endothelin-1 participates in the mechanism of disturbance and damage liver Ischemia median left lateral lobes induced for 60 min; it followed by reperfusion 24 hr. In some rats, antiserum or control serum without endothelin-1-blocking activity administered intravenously just before reperfusion. Rats were divided into three groups: an ischemia/reperfusion group that injected with serum, antiserum-treated sham-operated group. Endothelin-1 concentrations blood collected from suprahepatic vena cava measured use sandwich enzyme immunoassay. Index volume regional hepatic tissue index oxygenation assessed organ reflectance spectrophotometry system at 5 min 1, 2, hr The concentration started rise immediately onset basal values around 1 pg/ml reached value 6 reperfusion; maintained significantly high levels during period compared indicated lowered early phase group.(ABSTRACT TRUNCATED AT 250 WORDS)
