ABSTRACT
Previous studies suggest that treatment with icariin (ICA) combined Panax notoginseng saponins (PNS) improved behavior and cholinergic system disorders followed by amyloid beta-peptide(25-35) lateral ventricle injection in rats. The present study investigated whether administration of ICA + PNS had preventive therapeutic effects on bilateral common carotid arteries (CCA) occlusion-induced cerebral ischemia-reperfusion (IR) injury Male Sprague-Dawley rats were divided randomly as follows: sham-operated, i.g. vehicle, (5 mg/kg), (40 (2.5 20, 5 40 or 10 80 ergoloid mesylate a positive control (0.45 mg/kg) model Treatment was performed once day for 7 days prior to ischemia. subjected transient global IR induced CCA occlusion combination intraperitoneal sodium nitroprusside (2.0 then treated another 14 continuously. significantly the rat passive avoidance task step-down paradigms, spatial cognition eight-arm radial maze, concomitant an improvement blood viscosity. Increased lipid peroxidation brain after observed, MDA being 0.56 +/- 0.10 nmol/mg prot vs 0.48 0.06 vehicle (p < 0.05). 2.5 10, 40, mg/kg produced marked reduction level 0.46 0.06, 0.42 0.09 0.45 0.08 prot, respectively only 0.05, p 0.01). A decrease superoxide dismutase activity observed (the SOD 72.75 4.62 U/mg 80.97 6.06 control, prevented mediated fall 78.90 6.61 versus tended attenuate apoptosis hippocampal CA1 pyramidal neurons. Either alone did not obviously improve cognitive impairment (except reduced PNS-treatment). results indicated may ameliorate learning memory deficit viscosity protecting neurons from oxidative stress ischemic brain.
