Scalable Multiplexed Drug‐Combination Screening Platforms Using 3D Microtumor Model for Precision Medicine

ABSTRACT
Abstract Cancer heterogeneity is a notorious hallmark of this disease, and it desirable to tailor effective treatments for each individual patient. Drug combinations have been widely accepted in cancer treatment better therapeutic efficacy as compared single compound. However, experimental complexity cost grow exponentially with more target compounds under investigation. The primary challenge remains efficiently perform large‐scale drug combination screening using small number patient samples testing. Here, scalable, easy‐to‐use, high‐throughput scheme reported, which has the potential all possible pairwise arbitrary drugs multiple logarithmic mixing ratios. A “Christmas tree mixer” structure introduced generate concentration ratio between pairs, providing large range screening. three‐layer design special inlets arrangement facilitate simple loading process. As proof concept, an 8‐drug chip implemented, capable 172 different conditions over 1032 3D spheroids on chip. Using both cell lines patient‐derived cells, demonstrated precision medicine.
