A Comparative Transcriptomic Analysis Reveals Conserved Features of Stem Cell Pluripotency in Planarians and Mammals

ABSTRACT
Many long‐lived species of animals require the function adult stem cells throughout their lives. However, transcriptomes in invertebrates and vertebrates have not been compared, consequently, ancestral regulatory circuits that control cell populations remain poorly defined. In this study, we used data from high‐throughput RNA sequencing to compare pluripotent planarians with human mouse embryonic cells. From a stringently defined set 4,432 orthologs shared between planarians, mice humans, identified 123 conserved genes are ≥5‐fold differentially expressed all three species. Guided by gene set, RNAi screening discover novel regulators, which found affect cell‐associated functions tissue homeostasis, regeneration, maintenance. Examples disrupted these processes included TBL3, PSD12, TTC27, RACK1. analyses, concluded comparing diverse species, it is possible uncover factors biology. These results provide insights into comprised circuitry underlying self‐renewal pluripotency.
