Fine mapping of a Phytophthora-resistance gene RpsWY in soybean (Glycine max L.) by high-throughput genome-wide sequencing

ABSTRACT
Using a combination of phenotypic screening, genetic and statistical analyses, high-throughput genome-wide sequencing, we have finely mapped dominant Phytophthora resistance gene in soybean cultivar Wayao. root rot (PRR) caused by sojae is one the most important soil-borne diseases many soybean-production regions world. Identification resistant gene(s) incorporating them into elite varieties are an effective way for breeding to prevent from being harmed this disease. Two populations 191 F2 individuals 196 F7:8 recombinant inbred lines (RILs) were developed map Rps crossing susceptible Huachun 2 with Genetic analysis population indicated that PRR Wayao was controlled single gene, temporarily named RpsWY, which on chromosome 3. A high-density linkage bin constructed using 3469 recombination bins RILs explore candidate genes sequencing. The results genotypic showed RpsWY located 401 between 4466230 4502773 bp 3 through line 71 100 RILs. Four predicted (Glyma03g04350, Glyma03g04360, Glyma03g04370, Glyma03g04380) found at narrowed region 36.5 kb 401. These suggest resequencing method fine genes.
