Towards a broader view of the metabolome: untargeted profiling of soluble and bound polyphenols in plants

ABSTRACT
Phenylalanine (Phe) is a central precursor for numerous secondary plant metabolites with multitude of biological functions. Recent studies on the fungal disease Fusarium head blight in wheat showed Phe-derived defence to be induced presence pathogen. These also suggest partial incorporation into cell wall. To broaden view metabolome bound Phe derivatives, an existing approach using 13C-labelled as tracer was extended. The developed workflow consists three successive extractions acidified acetonitrile-methanol-water mixture remove soluble metabolites, followed by wall hydrolysis 4M aqueous NaOH, acidification HCl, and liquid-liquid extraction hydrolysate ethyl acetate. untargeted screening revealed 156 compounds 90 hydrolysed samples including known constituents like ferulic acid, coumaric tricin. Forty-nine were found exclusively hydrolysate. average cumulative yield 99.6%, range 91.8 100%. Repeatability coefficients variation protocol ranged from 10.5 25.9%, median 16.3%. demonstrate suitability proposed method typical metabolomics application, mock-treated graminearum-treated compared. study differences between hydrolysates two sample types, confirming differential under infection conditions.
