ABSTRACT
microRNA (miRNA) is a type of endogenous short-chain non-coding RNA with regulatory function found in eukaryotes, which involved the regulation variety cellular and biological processes. However, research on development cashmere goat secondary hair follicles still relatively scarce. In this study, small libraries mRNA 45 days, 55 65 75 days fetal skin goats were constructed, constructed sequenced using Illumina Hiseq4000, expression profiles miRNA obtained. The differentially expressed miRNAs mRNAs six control groups identified qRT-PCR experiment shows that sequencing results are accurate. Sixty-six related to follicle screened, used TargetScan miRanda predict 33 highly target genes. At same time, 664 GO enrichment KEGG pathway analysis performed. It was some genes consistent screening enriched Notch signaling pathway, TGF-β pathway. Therefore, miR-145-5p-DLL4, miR-27b-3p-DLL4, miR-30e-5p-DLL4, miR-193b-3p-TGF-β1, miR-181b-5p-NOTCH2, miR-103-3p-NOTCH2 network dual-luciferase reporter gene assay indicated there targeted relationship between chi-miR-30e-5p DLL4, will provide basis for molecular mechanism miRNA-mRNA goats.
