ABSTRACT
Abstract Exosomes exhibit great therapeutic potential in bone tissue engineering. The study aimed to investigate whether the exosomes derived from human adipose-derived stem cells (hADSCs-Exos) during different time-span of osteogenic differentiation could promote osteogenesis. appropriate concentrations hADSCs-Exos enhance proliferation, migration and osteogenesis were also examined. PKH67 labelled was used detect internalization ability hADSCs. abilities hADSCs after treatment with evaluated by Alizarin red staining (ARS). proliferation examined cell counting kit-8 wound healing assay, respectively. expression exosomal surface markers osteoblast-related protein assessed Western blot. PKH67-labelled internalized 4 h incubation. ARS showed that amount mineralized nodules Exo 1−14d group significantly higher than 15−28d group. capacity blot analysis treatment, ALP RUNX2 enhanced. Specially, 15 μg/mL upregulated other treated groups. Our findings suggest secreted induction for 1–14 days be efficiently induce Moreover, administration at promoted In conclusion, our research confirmed comprised may provide a new paradigm
