ABSTRACT
The cytomegalovirus (CMV) promoter is one of the most commonly used promoters for expression transgenes in mammalian cells. aim our study was to evaluate role methylation and upregulation CMV by irradiation chemotherapeutic agent cisplatin vivo using non-invasive fluorescence imaging.Murine fibrosarcoma LPB mammary carcinoma TS/A cells were stably transfected with plasmids encoding p21 promoter-driven green fluorescent protein (GFP) gene. Solid tumors induced subcutaneous injection tumor cells, while leg muscles transiently plasmid GFP under control promoter. Cells, tumors, legs treated either DNA inhibitor 5-azacytidine, irradiation, or cisplatin. determined a microplate reader vitro imaging vivo.Treatment 5-azacytidine (re)activated Furthermore, treatment resulted significant both vivo.Observed alterations activity limit usefulness this widely as constitutive On other hand, inducibility can be beneficially gene therapy when combined standard cancer treatment, such radiotherapy chemotherapy.
