ABSTRACT
Abstract Ageing is a complex biological process with variations among individuals, leading to the development of ageing clocks estimate age. Glycans, particularly in immunoglobulin G (IgG), have emerged as potential biomarkers ageing, changes glycosylation patterns correlating chronological For precision analysis, three different plasma pools were analysed over 26 days tetraplicates, 312 samples total. In short-term variability two cohorts analysed: AstraZeneca MFO cohort healthy individuals (median age 20) and 70 premenopausal Chinese women 22.5) monitored 3 months. Long-term analysis involved adult men aged 47 57, for 5 10 years, respectively. Samples collected every months weeks, IgG N -glycan followed standardized approach by isolating IgG, its subsequent denaturation deglycosylation glycan cleanup labelling. Capillary gel electrophoresis laser-induced fluorescence (CGE-LIF) ultra-performance liquid chromatography analyses employed profiling. Statistical normalization, batch correction, linear mixed models assess time effects on derived traits. The intermediate results consistently exhibited very low coefficient variation values across all test samples. This consistent pattern underscores high level inherent CGE method analysing clock ageing. did not show any statistically significant trends, whereas menstrual cycle trends digalactosylated (G2), agalactosylated (G0) fucosylation (F). These attributed cycle. stability identified enduring age-related both subjects, showing positive effect G0 bisected -acetylglucosamine, well negative G2 sialylation, aligning earlier findings.
