ABSTRACT
Microglia play an important role in neuronal protection and damage. However, the molecular cellular relationship between microglia neurons is unclear. We carried out a prospective study to detect that activation of BV2 induced PC12 cell apoptosis vitro through TLR4/adapter protein myeloid differentiation factor 88 (MyD88)/nuclear factor-κB (NF-κB) signaling pathway. were treated with different concentrations LPS for 24 h. Western blot was utilized expression TLR4 downstream The level inflammatory mediator quantified using specific ELISA kit. supernatant 10 μg/ml LPS-treated cells used as conditioned medium (CM). co-culture CM Cell viability determined by MTT assay tested flow cytometry. 10, 20, or 30 TLR4, MyD88, NF-κB significantly increased. When co-cultured h, decreased. up-regulated Bax down-regulated Bcl-2 cells. pretreated interleukin-1 receptor antagonist (IL-1RA) min, alleviated CM-induced apoptosis. These results suggest activated triggered TLR4/MyD88/NF-κB pathway release IL-1β could participate injury.
