Wogonin attenuates inflammation and oxidative stress in lipopolysaccharide-induced mastitis by inhibiting Akt/NF-κB pathway and activating the Nrf2/HO-1 signaling

ABSTRACT
Mastitis is a disease involved in inflammation of breast which affects human and animals. Wogonin one bioactive compound from many Chinese herbal medicines, have multiple properties, including anti-inflammatory activity. However, the roles wogonin mastitis progression are largely undefined. models were established using LPS-treated mice mammary epithelial cells (MECs). Infiltration inflammatory was analyzed by hematoxylin-eosin staining myeloperoxidase (MPO) Inflammatory cytokine (TNF-α IL-1β) levels detected via ELISA. The phosphorylation total Akt NF-κB content Nrf2 HO-1 measured western blot. Cell viability examined CCK-8 assay. Oxidative stress assessed ROS generation MDA, GSH, SOD. attenuated LPS-induced infiltration cells, increase MPO activity TNF-α IL-1β, activation Akt/NF-κB pathway murine gland tissues, promoted Nrf2/HO-1 signaling. did not affect MEC viability, but mitigated MECs reducing IL-1β levels. relieved oxidative through decreasing MDA level increasing GSH SOD repressed increased signaling activation. Activated or inactivation reversed suppressive effects on MECs. mitigates suppressing activating pathway, indicating therapeutic potential mastitis.
