ABSTRACT
This study investigated resveratrol impact on oral squamous cell carcinoma (OSCC) via the NORAD/IGF2BP2/PDK1 pathway. CAL-27, SCC-25, and KB lines were used to evaluate proliferation, cycle arrest, protein expression. Key molecular markers assessed using Western blot, RNA interference, functional assays. Resveratrol inhibited growth of KB, SCC-25 cancer in a dose-dependent manner, with IC50 values 70, 145, 125 μg/mL, respectively (P < 0.01). In CAL-27 cells, 50 μg/mL induced G2/M arrest 0.05); 100 caused S phase Thirteen proteins changed significantly: cPKCα Notch4 upregulated, while p-ERK, p-PDK1, p-Cdc2, p-RB, NORAD, IGF2BP2, CDK2, Cdc2P34, Cyclin E, 14-3-3beta, XIAP downregulated. si-NORAD groups showed lower proliferation than control. IGF2BP2 silencing reduced 69.13% HSC3 74.01% 0.001) decreased invasion 72.85% 52.44% 0.001). PDK1 overexpression enhanced migration hypopharyngeal cells. inhibits OSCC particularly It affects pathways, altering expression causing arrest.
