ABSTRACT
This study investigated the anticancer effect and mechanism of salinomycin, a selective inhibitor cancer stem cell, on human ovarian cell line OV2008 in vitro vivo. The growth inhibitory salinomycin was determined by measuring viability using resazurin reduction assay. Apoptotic nuclear morphology visualized 4,6-diamino-2-phenylindole staining technique. percentages apoptotic cells cycle parameters were detected flow cytometry. activity p38 mitogen-activated protein kinase (p38 MAPK) analyzed Bio-Plex phosphoprotein In vivo assayed through tumor growth. Salinomycin caused concentration- (0.01-200 μM) time-dependent (24-72 h) effects OV2008. Cell observations showed that salinomycin-treated displayed typical characteristics. significantly increased OV2008, showing manner. There no arrest G1/G0, S, G2/M phases between control cells. also enhanced phosphorylation MAPK. Moreover, inhibited xenograft tumors. exhibited significant inhibition induction apoptosis data suggested salinomycin-induced might be associated with activating MAPK merits further investigations.
