Inhibition of human 67-kDa laminin receptor sensitizes multidrug resistance colon cancer cell line SW480 for apoptosis induction

ABSTRACT
The adhesion mediated drug resistance in cancer cells resulted from of the extracellular matrix is a major cause for multidrug (MDR) and leads chemotherapeutic failure colon cancer. In this study, we explored role 67-kDa laminin receptor (67LR) cells. SiRNA-mediated knockdown 67LR decreased cell when laminins were applied. Moreover, increased expression pro-apoptotic gene Bax but inhibited anti-apoptotic Bcl-2. Enhanced apoptosis was observed siRNA-transfected SW480 treated with doxorubicin induction. Furthermore, MTT assay revealed that IC50 toward to reduced after knockdown, indicating there significant increase sensitivity cell. conclusion, our study demonstrated plays considerable development MDR.
