A new insight on copper: Promotion of collagen synthesis and myofiber growth and development in juvenile grass carp (Ctenopharyngodon idella)

ABSTRACT
Copper (Cu) is a trace element, essential for fish growth. In the current study, in addition to growth performance, we first explored effects of Cu on collagen synthesis and myofiber development juvenile grass carp (Ctenopharyngodon idella). A total 1080 (11.16 ± 0.01 g) were randomly divided into 6 treatments (3 replicates per treatment) receive five doses organic Cu, which citrate (CuCit) at 0.99 (basal diet), 2.19, 4.06, 6.15, 8.07 mg/kg, one dose inorganic (CuSO4·5H2O 3.15 mg/kg), 9 weeks. The results showed appropriate level (4.06 mg/kg) enhanced improved nutritional status, downregulated Cu-transporting ATPase 1 mRNA levels hepatopancreas, intestine, muscle carp. Meanwhile, content was increased after intake, probably due following pathways: (1) activating CTGF/TGF-β1/Smads signaling pathway regulate transcription; (2) upregulating La ribonucleoprotein domain family (LARP6) translation initiation; (3) increasing proline hydroxylase, lysine oxidase activities posttranslational modifications. addition, optimal group diameters frequency myofibers with diameter >50 μm, might be associated upregulation cyclin B, D, E, proliferating cell nuclear antigen, myogenic determining factor (MyoD), 5, myogenin (MyoG), regulatory 4 myosin heavy chain (MyHC) downregulation myostatin levels, protein MyoD, MyoG MyHC muscle. Finally, based percentage weight gain (PWG), serum ceruloplasmin (Cp) activity muscle, requirements determined as 4.74, 4.37 4.62 mg/kg diet (CuCit source) carp, respectively. Based PWG Cp activity, compared CuSO4·5H2O, efficacy CuCit 131.80% 115.38%, Our findings provide new insights supplementation promote fish, help improve overall productivity aquaculture.
