ABSTRACT
EGF receptor (EGFR) is a critical signaling node throughout life. However, it has not been possible to directly visualize endogenous Egfr in mice. Using CRISPR/Cas9 genome editing, we appended fluorescent reporter the C terminus of Egfr. Homozygous mice appear normal and EGFR intact vitro vivo. We detect distinct patterns expression progenitor differentiated compartments embryonic adult Systemic delivery or amphiregulin results markedly different internalization trafficking hepatocytes. In intestine, localizes crypt rather than villus compartment, higher adjacent epithelium intestinal tumors, following colonic injury appears cell populations stroma. This reporter, under control its regulatory elements, enables vivo monitoring dynamics localization disease states.
