Identification of hematopoietic stem cells residing in the meninges of adult mice at steady state

ABSTRACT
Steady-state extramedullary hematopoiesis during adulthood is an emerging field of great interest. The meninges contain both innate and adaptive immune cells, which provide immunosurveillance the central nervous system (CNS). Hematopoietic progenitors that give rise to meningeal cells remain elusive. Here, we report steady-state adult mice host hematopoietic stem (HSCs), as defined by long-term, efficient, multi-lineage reconstitution self-renewal capacity in meninges, blood, spleen, bone marrow sublethally irradiated recipients. HSCs lodge after birth with local expression pro-hematopoietic niche factors. Meningeal are locally maintained homeostasis get replenished from blood only when resident pool reduced. With a tissue-specific profile, can CNS constant supply leukocytes more adapted microenvironment.
