Rapid detection of human influenza A viruses by HFman probe-based loop-mediated isothermal amplification assays

ABSTRACT
Since China abandoned the zero-COVID policy at end of 2022, a wave severe Flu pandemic emerged in China. Rapid and accurate diagnosis Influenza A virus (IAV) is critical for clinical management therapeutic decision-making patients with fever. Here, we reported novel IAV HF-LAMP assay, which can be performed purified RNA or directly using samples. The assays samples have high sensitivity limit detection (LOD) 9.6 copies/reaction, 9900 copies/mL, short sample-to-answer times 36 50 min, respectively. Both showed specificity significantly higher rate than rapid antigen (RAD) assays. Furthermore, found vast majority (91.2 %) children fever during were infected by IAV, current infection has very narrow detectable window. IVA will provide robust tools to facilitate early future seasonal influenza epidemics.
