Investigating the mechanism of action of Yanghe Pingchuan Granule in the treatment of bronchial asthma based on bioinformatics and experimental validation

ABSTRACT
Yanghe Pingchuan Granule (YPG) is a patented Chinese medicine developed independently by the Anhui Provincial Hospital of Traditional Medicine. For many years, it has been used for treatment asthma with remarkable clinical effects. However, composition YPG complex, and its potential active ingredients mechanism action are unknown. In this study, we investigated in through combination bioinformatics vivo experimental validation. We searched compounds targets from multiple databases obtained common targets. Subsequently, protein-protein interaction (PPI) network compound disease was constructed using protein database Gene Ontology (GO) analysis Kyoto Encyclopedia Genes Genomes (KEGG) pathway enrichment analysis. Finally, hematoxylin eosin (H&E) staining, Masson enzyme-linked immunosorbent assay (ELISA) analysis, immunofluorescence (IF) experiments, Western blot (WB) experiments were performed to verify possible treatment. 72 318 drug target genes that overlap asthma. Serine/threonine-protein kinase (AKT1), tumor p53 (TP53), necrosis factor (TNF), interleukin (IL)-6, IL-1β, vascular endothelial growth factor-A (VEGFA), prostaglandin-endoperoxide synthase 2 (PTGS2), caspase-3 (CASP3), mitogen-activated 3 (MAPK3) epidermal receptor (EGFR) most relevant PPI network. KEGG showed high number enriched nuclear kappa-B (NF-κB) signaling pathway. Animal confirmed reduced inflammatory cell infiltration down-regulated expression ovalbumin-induced factors. Furthermore, decreased NFĸB1, kappa B subunit beta (IKBKB), (VEGF), (VEGFR2) lung tissue. positive effect on interfering may significantly inhibit follicle-induced response NF-ĸB
