Phytochemical composition and safety of Vernonia Amygdalina ethanolic extract with anti-colon cancer properties

ABSTRACT
This study evaluates the therapeutic efficacy and safety of Vernonia Amygdalina extract (VAEE) in treating colorectal cancer (CRC) induced by azoxymethane/dextran sulfate sodium (AOM/DSS) mice to elucidate underlying mechanisms. The utilized a mouse model CRC assess impact VAEE on pathological markers, inflammatory mediators, oxidative stress. Phytochemical profiling was conducted using LC-HRMS, focusing phenolic compounds. extract's antioxidant activity quantified, its profile verified through acute toxicity assessments. significantly mitigated AOM/DSS-induced reduction colon length weight loss dose-dependent manner. treatment decreased pro-inflammatory cytokines (IL-6, IL-1β, TNF-α) while increasing anti-inflammatory cytokine IL-10. also demonstrated strong did not adversely affect blood, kidney, or liver biomarkers, confirming non-toxicity. Protein-protein interaction analysis suggested VAEE's role inhibiting including COX-2, IL-18, NF-kB. Molecular docking results supported affinity for these proteins, indicating potential as COX-2 inhibitor.VAEE exhibits potent anti-inflammatory, antioxidative, anticarcinogenic properties against CRC, with favorable profile.
