ABSTRACT
We aimed to assess whether β-amyloid (PIB) and tau (AV1451) PET predict longitudinal atrophy in patients with AD. A group of 10 fulfilling NIA-AA criteria for AD dementia likely due were included (age = 63 ± 9, MMSE 24 4 at baseline). All underwent i) a baseline visit structural MRI imaging both AV1451 PIB, ii) follow-up (time between MRIs 1.06 0.15 years). Structural images preprocessed using SPM12's registration pipeline obtain voxelwise maps showing areas local contractions expansions (i.e. Jacobians, see figure 1A). Relationships data (Standardized Uptake Value Ratio (SUVR) normalized cerebellar gray matter) subsequent assessed two complementary ways. First, global cortical value was extracted each patient modality (figure 1B) correlations computed the level. Second, we individual level evaluate similarity uptake map 1C). Using measures 2A), significant association observed steeper (lower Jacobians) higher SUVR (rho -0.76, p 0.02) but not PIB-SUVR 0.13, 0.73), Fig 2a. Similarly, correlation analyses revealed that resembled AV1451-PET (median rho -0.57), i.e. voxels AV1451-uptake showed rate atrophy. Correlation PIB minimal -0.14) significantly different from (Wilcoxon signed-rank test: p=0.004, Figure 2B). These results support hypothesis neurodegeneration is more closely related than pathology, further suggests pathology precedes drives locally. Our suggest potential prognostic role predicting trajectories. From clinical perspective, our could have major utility short-term outcomes patients.
