ABSTRACT
The R-loop is a naturally formed three-strand nucleic acid structure that recently has been reported to participate in multiple biological processes and helped answer some previously unexplained scientific questions. Meiosis process involves chromatin-related events such as DNA double-stranded breaks (DSB) formation, repairing transcriptional dynamics.Explore the regulatory roles physiological functions of R-loops mammalian meiosis process.In our study, using genome-wide S9.6 CUT & Tag seq, we first mapped genomic distribution dynamic changes during meiotic mice, from spermatogonia secondary spermatocytes. And further explore role conditions by constructing conditional knockout mice Rnaseh1, which deleted endonuclease before entry.R-loop predominantly distributes at promoter-related regions varies across different stages. By joint analysis with corresponding transcriptome, found was closely related transcription process. high frequency cells usually accompanied activity, verified this leptotene/zygotene pachytene transition Moreover, lack RNase H1 caused sterility male accumulation abnormal DSB repair Further showed stages influenced regulation stage.The mutual plays an essential spermatogenesis. also important for normal meiosis.
