Engineered short forms of perlecan enhance angiogenesis by potentiating growth factor signalling

ABSTRACT
Growth factors are key molecules involved in angiogenesis, a process critical for tissue repair and regeneration. Despite the potential of growth factor delivery to stimulate limited clinical success has been achieved with this approach. interact extracellular matrix (ECM), particularly heparan sulphate (HS), bind potentiate their signalling. Here we show that engineered short forms perlecan, major HS proteoglycan vascular ECM, signal angiogenic factors, including fibroblast 2 endothelial factor-A. We also perlecan delivered porous chitosan biomaterial scaffolds promote angiogenesis rat full thickness dermal wound model, fusion domains I V leading superior vascularisation compared native or alone. Together, study demonstrates as next generation therapies which exert biological activity via potentiation factors.
