ABSTRACT
Non melanoma skin cancers are common neoplasms worldwide. In India, squamous cell carcinoma (SCC), is the most prevalent disorder and its incidence rises quickly with cumulative exposure to sun. Numerous techniques available for SCC but reversion metastasis concern that needs effective safe strategies control. With this in view, study was planned investigate activity of Bakuchiol (Bak), traditionally used various countries curing ailments mechanism action unexplored. our study, we explored anti-proliferative, pro-apoptotic anti-inflammatory potential Bak toward human (A431) line. The pure compound isolated from plant Psoralea corylifolia characterized using NMR, HRMS FTIR. To explore their bioefficacy, different vitro assays were performed against A431 have molecular insights, RT-qPCR investigation done analyzed expression level inflammatory markers (TLR 9, IFN β, IL 23, JAK 3 STAT 3). results showed growth inhibitory effect on cells after treatment dose-dependent way. understand mode death, initially under phase-contrast, fluorescence scanning electron microscope characteristics apoptosis. Furthermore, cycle studies a flow cytometer carried out which increased ROS, reduced MMP arrested at G0/G1 phase treated further strengthening induction Moreover, analysis indicated downregulation Bak-treated confirmed therapeutic role. docking also has perfect scaffold can complete pharmacophoric need JAK3 kinase inhibition. A critical points towards role ameliorating along apoptosis by regulating variable markers.
