ABSTRACT
Reflecting its pleiotropic functions, Polo-like kinase 1 (PLK1) localizes to various sub-cellular structures during mitosis. At kinetochores, PLK1 contributes microtubule attachments and mitotic checkpoint signaling. Previous studies identified a wealth of potential receptors at as well requirements for kinases, including BUB1, Aurora B, itself. Here, we combine ectopic localization, in vitro reconstitution, kinetochore localization demonstrate that most likely all the is recruited through BUB1 outer centromeric protein U (CENP-U) inner kinetochore. CENP-U share constellation sequence motifs consisting putative PP2A-docking motif two neighboring PLK1-docking sites, which, contingent on priming phosphorylation by cyclin-dependent itself, bind promote dimerization. Our results rationalize previous observations describe unifying mechanism recruitment human kinetochores.
