ABSTRACT
Hemorrhagic fever with renal syndrome (HFRS), caused by Orthohantaviruses, occupies one of the leading places among natural focal human diseases, for which there are no modern accurate and highly sensitive diagnostic methods. To improve this situation, a better understanding Hantavirus pathogenesis HFRS is required. Determination expression level exosomal microRNAs (miRNAs) in serum/plasma patients makes them potential biomarkers diagnosing predicting HFRS. The purpose study was to analyze miRNA-146a, miRNA-126, miRNA-218, miRNA-410, miRNA-503 miRNA-155 at different stages (fever stage, polyuric stage convalescence stage) severity course disease. The moderate group included 105 patients, severe 99 complications 84 patients. Blood samples from molecular genetic analysis were collected three times: during initial febrile period (days 1-4 illness), 15-22 disease), convalescence. Total RNA isolation performed using exoRNeasy Midi Kit (Qiagen, Germany). Quantitative real-time PCR (qRT-PCR) miRCURY LNA SYBR Green Germany) LightCycler96 product detection system (Roche, Switzerland). When comparing miRNAs groups disease, statistically significant increase miRNA-146a revealed (Fold change 2.694; p = 0.0022) compared disease form, as well an form 1.861; 0.0492; Fold 1.976; 0.001, respectively). Comparative other various did not reveal any results (P > 0.05).MiRNA-155 may be promising prognostic However, further investigations needed evaluate changes network genes that can targets studied order elucidate mechanisms influence occurrence development
