ABSTRACT
The pathogenesis of depression remains largely unknown. Accumulating evidence demonstrates the existence a complex relationship between gut microbiome composition and brain functions. Jia Wei Xiao Yao San (JWXYS) is considered potential antidepressant. However, pharmacological mechanisms JWXYS have not yet been clarified. This study aimed to explore effects on chronic stress-induced depression-like behaviors in mice. A restraint stress mouse model was established. administered, responses these mice treatment were evaluated through several behavioral tests. activity astrocytes microglia detected by specific fluorescent labels. Inflammatory cytokines quantified intestinal cerebral tissues. An integrated approach with full-length 16S rRNA sequencing different types untargeted metabolomics conducted investigate at species level, metabolic functions, JWXYS. We found that symptoms associated relative abundance Lactobacillus animalis. After treatment, Ileibacterium valens enzymes potentially involved purine metabolism also described. activation negatively correlated L. Combined network analysis, targets predicted based focused metabolism, which enriched from metabolites regulated Our suggests animalis increases I. relation positively amygdala.
