Pregnane alkaloids with BRD4 inhibitory and cytotoxic activities from Pachysandra terminalis

Abstract Three new pregnane alkaloids, pactermines A – C (1 – 3), were obtained from the whole plant of Pachysandra terminalis. The structures of these alkaloids were elucidated by comprehensive spectroscopic methods including 1D, 2D NMR, MS and IR data analysis. Pactermines A and B showed moderate bromodomain-containing protein 4 (BRD4) inhibitory activities with IC50 values of 10.1 and 19.4 μM. In addition, pactermines A and B exhibited cytotoxic activities against three tumor cell lines with IC50 values ranging from 5.3–57.4 μM.
