ABSTRACT
The research objective was to develop a thermostable vaccine against peste des petits ruminants (PPR), morbilliviral disease of small targeted for eradication that is major constraint on the livelihoods rural poor throughout much Africa and Asia. Although existing PPR vaccines provide life-long immunity, they require continuous refrigeration. This limits their utility in developing countries. Methods lyophilization related morbillivirus, rinderpest (RP), resulted could be used field up 30days without refrigeration which contribution global RP completed 2011. present applied method attenuated Nigeria 75/1 strain, measured thermostability accelerated stability tests (AST) at 37°C. shelf-life determined as time vial retained minimum dose required 25-dose presentation specified temperature. A lactalbumin hydrolysate sucrose (LS) stabilizer compared stabilizers based trehalose. produced using Xerovac drying AST. LS evaluated AST 37, 45 56°C an Arrhenius plot constructed estimation temperatures not tested. Vaccines were most stable. preparation 22.2days three batches had shelf-lives 37°C 177.6, 105.0 148.9days, respectively, At 56°C, 13.7days. projected half-life 25°C 1.3years. sufficient use cold chain will greatly facilitate delivery vaccination PPR.
