ABSTRACT
Dexamethasone (dex) is a glucocorticoid that mainstay for the treatment of inflammatory pathologies, including immunotherapy-associated toxicities, yet specific impact dex on activity CAR T cells not fully understood. We assessed whether given ex vivo or as an adjuvant in with impacted phenotype function cells. demonstrated cell expansion and were inhibited by dex. confirmed this observation using multiple constructs tumor models, suggesting general phenomenon. Moreover, we determined upregulated interleukin-7 receptor α increased expression genes involved activation, migration, persistence when supplemented vivo. Direct delivery IL-7 into tumor-bearing mice resulted adoptively transferred complete regression. Overall, our studies provide insight use to enhance therapy represent potential novel strategies augmenting during production well following infusion patients.
