ABSTRACT
Herein, a pipette-tip-enabled digital nucleic acid analyzer for high-performance COVID-19 testing is demonstrated. This achieved by loop-mediated isothermal amplification (digital LAMP or dLAMP) using common laboratory equipment and materials. It shown that simply fixing glass capillary inside conventional pipette tips enables the generation of monodisperse, water-in-oil microdroplets with benchtop centrifugation. LAMP, ORF1a/b gene, standard test region screening, can be amplified without thermal cycler. The allows counting fluorescent so Poisson analysis performed to allow quantification limit detection 1 order magnitude better than those nondigital techniques comparable commercial dLAMP platforms. envisioned this work will inspire studies on ultrasensitive analyzers demanding both sensitivity accessibility, which pivotal their large-scale applications.
