ABSTRACT
Lacto-N-biose (LNB) is a member of the human milk oligosaccharide (HMO) family and synthesized via an enzymatic reaction in vitro with N-acetylglucosamine (GlcNAc) cofactors. In this study, LNB was using cell factory for first time. First, three modules were constructed Kluyveromyces lactis producing from lactose GlcNAc without addition Second, de novo pathway K. adding GlcNAc. Finally, transcriptional switch introduced into to reprogram its metabolic network improving flux GlcNAc-6-P pathway. Subsequently, final yield 10.41 g/L, similar salvage yield, achieved through The engineered provides promising technology platform industrial scale production LNB.
