Chemoprevention of Colorectal Cancer by Artocarpin, a Dietary Phytochemical from Artocarpus heterophyllus

ABSTRACT
Artocarpus heterophyllus is an evergreen tree distributed in tropical regions, and its fruit (jackfruit) well-known as the world's largest tree-borne fruit. Although A. has been widely used folk medicines against inflammation, potential cancer chemoprevention remains unclear. Herein we identified artocarpin from a promising colorectal chemopreventive agent by targeting Akt kinase. Phenotypically, exhibited selective cytotoxicity human colon cells. Artocarpin impaired anchorage-independent growth capability, suppressed cell growth, induced G1 phase cycle arrest which was followed apoptotic well autophagic death. Mechanistic studies revealed that directly targeted 1 2 kinase activity evidenced vitro assay, ex vivo binding assay downstream cellular signal transduction. Importantly, oral administration of attenuated colitis-associated tumorigenesis mice. Taken together, artocarpin, bioactive component heterophyllus, might merit investigation agent.
