ABSTRACT
Gossypol, commonly found in cotton seeds, is hazardous to male reproductive physiology. Although several studies have indicated the toxicity of gossypol human and animal reproduction, mechanism action testes has not yet been elucidated. In present study, we investigated effects normal mouse testis cells, TM3 TM4 gossypol-treated C57BL/6 mice. We confirmed antiproliferative using cell viability assays, with PCNA as a proliferation marker, cycle analysis. also verified mitochondrial dysfunction Ca2+ dysregulation cytosol JC-1 Fluo-4 dyes. To confirm cellular signaling mechanisms lines, performed Western blot analysis assess changes MAPK PI3K/Akt signal transduction, their pharmacological inhibitors. Moreover, screened mRNA expression genes involved spermatogenesis steroidogenesis cells. localization regulating function untreated mice testes. Collectively, suggest that induces negative on by reducing viability, membrane potential, development-related vitro vivo well modulating PI3K pathways.
