ApoE-Dependent Protective Effects of Sesamol on High-Fat Diet-Induced Behavioral Disorders: Regulation of the Microbiome-Gut–Brain Axis

ABSTRACT
Sesamol, an antioxidant lignan from sesame oil, possesses neuroprotective bioactivities. The present work was aimed to elucidate the systemic protective effects of sesamol on cognitive deficits and determine possible link between gut brain. Wildtype ApoE–/– mice were treated with a high-fat diet (0.05%, w/v, in drinking water) for 10 weeks. Behavioral tests including Morris-water maze, Y-maze, elevated plus maze indicated that could only improve anxiety behaviors wildtype. Consistently, improved synapse ultrastructure inhibited Aβ accumulation ApoE-dependent manner. Moreover, prevented dietary-induced barrier damages inflammation. Sesamol also reshaped microbiome generation microbial metabolites short-chain fatty acids. To summarize, this study revealed mechanism might be ApoE-dependent, its beneficial microbiota/metabolites translated into neurodegenerative diseases treatment.
