ABSTRACT
While Lck has been widely recognized to play a pivotal role in the initiation of T cell receptor (TCR) signaling pathway, an understanding precise regulation cells upon TCR activation remains elusive. Investigation protein–protein interaction (PPI) using proximity labeling techniques such as TurboID potential provide valuable molecular insights into regulatory networks. By expressing Lck-TurboID Jurkat cells, we have uncovered dynamic, short-range protein network 30 min stimulation. In this novel application TurboID, detected 27 early signaling-induced Lck-proximal interactors living including known and interactors, validating discovery power tool. Our results revealed previously unappreciated PPI which may be associated with cytoskeletal rearrangement, ubiquitination proteins, mitogen-activated kinase cascade, coalescence LAT signalosome, formation immunological synapse. study, demonstrated for first time immune that can utilized unveil dynamics at scale consistent signaling.
