ABSTRACT
Nanopore analysis relies on ensemble averaging of translocation signals obtained from numerous molecules, requiring a relatively high sample concentration and long turnaround time the to results. The recapture subsequent re-reading same molecule is promising alternative that enriches signal information single molecule. Here, we describe how an asymmetric nanopore improves molecular ping-pong by promoting in trans reservoir. We also demonstrate could be improved linking target DNA carrier reduce diffusion, thereby achieving over 100 events. Using this methodology, its use accurately resolving nanostructure motifs along scaffold through repeated detection. Our method offers novel insights into control polymer dynamics within confinement opens avenues for development high-fidelity detection platform.
