ABSTRACT
Bone repair and regeneration process is markedly impaired in diabetes mellitus (DM) that affects hundreds of millions people worldwide. As a chronic inflammatory disease, DM creates proinflammatory microenvironment defective sites. Most the studies on DM-associated bone regeneration, however, neglect importance immunomodulation under condition adopt same approaches to normal healing, leading limited healing. In this study, we developed unique bioinspired injectable microsphere as an osteoimmunomodulatory biomaterial modulates macrophages create prohealing condition. The was self-assembled with heparin-modified gelatin nanofibers, interleukin 4 (IL4) incorporated into nanofibrous (NHG-MS). IL4 has binding domains heparin, heparin stabilizes cytokine, protects it from denaturation degradation, subsequently prolongs its sustained release modulate macrophage polarization. IL4-loaded NHG-MS switched M1 M2 phenotype, recovered M2/M1 ratio level, efficiently resolved inflammation, ultimately enhanced osteoblastic differentiation regeneration. development biomaterials harness power for immunomodulation, therefore, novel promising strategy enhance
