Analysis of Unamplified 16S rRNA of Ammonia-Oxidizing Bacteria in Activated Sludge by Spectrophotometry Using Gold Nanoprobes

ABSTRACT
The identification of the bacteria contributing to water purification processes in these systems is crucial for development and optimization wastewater treatment by engineers. We developed a simple assay colorimetric quantification bacterial 16S rRNA functional extracted from samples without reverse transcription RNA or quantitative polymerase chain reaction (qPCR). heat test sample significantly reduced time (<3 min) improved sensitivity. Following optimization, calibration curve ammonia-oxidizing (AOB) β subdivision class Proteobacteria was created using activated sludge samples. linear above 1.9 × 106 copies/mL AOB, which 2 orders magnitude lower than that our previous studies. Finally, we determined abundance AOB compared results obtained transcription-qPCR. There some correlation concentrations both methods. Thus, have successfully an innovative tool quantify concentration processes.
