ABSTRACT
Abstract Genetic circuits and metabolic pathways can be reengineered to allow organisms process signals manufacture useful chemicals. However, their functions currently rely on organism-specific regulatory parts, fragmenting synthetic biology engineering into host-specific domains. To unify efforts, here we have engineered a cross-species expression resource that enables reuse the same genetic while functioning similarly across diverse organisms. Our system combines mixed feedback control loops translation autonomously self-regulate of an orthogonal polymerase without promoters, achieving nontoxic tuneable gene in Gram-positive Gram-negative bacteria. Combining 50 characterized variants with mechanistic modelling, show how resource’s dynamics, capacity toxicity are controlled by loops’ architecture strengths. We also demonstrate one application reusing parts express biosynthesis pathway both model non-model hosts.
