ABSTRACT
Chondroitin polymerizing factor (CHPF) is an important glycosyltransferase involved in the biosynthesis of chondroitin sulfate. However, relationship between CHPF and gastric cancer has not been fully investigated. expression tissues was detected by immunohistochemistry correlated with patient prognosis. Cultured cells human epithelial cell line GES1 were used to investigate effects shCHPF shE2F1 on development progression MTT, western blotting, flow cytometry analysis apoptosis, colony formation, transwell xenograft mouse models, vitro vivo. In tissues, found be significantly upregulated, its tumor infiltration advanced stage shorter survival cancer. may promote regulating proliferation, apoptosis migration, while knockdown induced opposite effects. Moreover, results from vivo experiments demonstrated that growth suppressed knockdown. Additionally, E2F1 identified as a potential downstream target regulation cancer, decreased CHPF-induced promotion Mechanistic study revealed regulate through affecting UBE2T-mediated ubiquitination. This showed, for first time, prognostic indicator promoter whose function likely carried out E2F1.
