ABSTRACT
Abstract Turn-on fluorescence imaging is routinely studied; however, turn-on chemiluminescence has been rarely explored for in vivo imaging. Herein, we report the design and validation of probe ADLumin-1 as a amyloid beta (Aβ) species. Two-photon indicates that can efficiently cross blood–brain barrier provides excellent contrast Aβ plaques cerebral angiopathy. In brain shows signal from 5-month-old transgenic 5xFAD mice 1.80-fold higher than age-matched wild-type mice. Moreover, demonstrate it feasible to further d ually- mplify s ignal via c hemiluminescence r esonance e nergy t ransfer (DAS-CRET) using two non-conjugated smart probes (ADLumin-1 CRANAD-3) solutions, homogenates, whole Our results show DAS-CRET provide 2.25-fold margin between wild type We believe our strategy could be extended other aggregating-prone proteins.
