Unequivocal identification of intracellular aluminium adjuvant in a monocytic THP-1 cell line

ABSTRACT
Abstract Aluminium-based adjuvants (ABA) are the predominant used in human vaccinations. While a consensus is yet to be reached on aetiology of biological activities ABA several studies have identified shape, crystallinity and size as critical factors affecting their adjuvanticity. In spite recent advances, fate following administration remains unclear. Few if any demonstrated unequivocal presence intracellular ABA. Herein we demonstrate for first time identification within monocytic T helper 1 (THP-1) cell line, using lumogallion fluorescent molecular probe aluminium. Use these new methods revealed that particulate was only found cytoplasm. Transmission electron microscopy were contained vesicle-like structures approximately 0.5–1 μm diameter.
