ABSTRACT
Abstract The product template (PT) domains, specifically in fungal non-reducing polyketide synthases (NR-PKSs), mediate the regioselective cyclization of polyketides dominating final structures. However, up to date, systematic knowledge about PT domains has been insufficient. In present study, relationships between sequences, structures and functions were analyzed with 661 NR-PKS sequences. Based on phylogenetic analysis, classified into prominent eight groups (I–VIII) corresponding representative compounds regioselectivity (C2-C7, C4-C9 C6-C11). Most cavity lining residue (CLR) sites all common, while regional CLR site mutations resulted appearance finger-like regions different orientation. volumes shapes, even catalytic dyad positions characteristic compound sizes. conservative residues sequences responsible for evolution key differentiations functions. above findings may help better understand mechanisms predict structural types aromatic products.
