ABSTRACT
Significance Functional analysis of noncoding genomic regulatory elements, which harbor the majority common human disease and trait associations, is complicated by their cellular context sensitivity. We developed Big-IN, a method for rewriting large segments mammalian genomes, including full genes surrounding elements. demonstrate flexible verification pipeline to identify correctly engineered cells. expect Big-IN will enable technologies synthesis assembly DNAs catalyze synthetic approach genomics.
